اثر تمرین استقامتی بر بیان ژن های هموباکس سی هشت (HOXC8) و سی نه (HOXC9) در بافت چربی موش های صحرایی نر نژاد ویستار
پروتئینهای هموباکس سی هشت(HOXC) و سی نه (HOXC9) پروتئین هایی هستند که در نمو بافت چربی نقش دارند. هدف از این پژوهش، بررسی اثر تمرین استقامتی بر بیان ژن های هموباکس سی هشت و سی نه در بافت چربی سفید زیر پوستی بود. برای این منظور، 16 موش (سن هشت نه هفته با میانگین وزن 220 گرم) صحرایی نژاد ویستار به صورت تصادفی به دو گروه؛ 1)کنترل (هشت سر) و 2) تمرین استقامتی (هشت سر) تقسیم شدند. موشهای گروه تمرینی، به مدت هشت هفته، هفتهای پنج جلسه، تحت تمرین استقامتی تداومی بر روی نوارگردان (با میانگین شدت 25 متر در دقیقه و با میانگین مدت 25 دقیقه) قرار گرفتند. برای اندازهگیری بیان نسبی ژنهای هدف در بافت چربی زیرپوستی از روش ریل تایم (Real Time–PCR)استفاده شد. دادهها نشان داد که بیان نسبی ژن پروتئین غیر جفت کننده یک (UCP1) در گروه استقامتی در مقایسه با گروه کنترل به صورت معناداری بیشتر بود (P=0.018). با این حال، بیان نسبی ژن های هموباکس سی هشت و سی نه در گروه استقامتی در مقایسه با گروه کنترل از نظر آماری تفاوت معناداری نداشت (P hoxc9=0.36; Phoxc9=0.52). یافتههای این مطالعه نشان داد که تمرین استقامتی تغییری در بیان ژنهای هموباکس سی هشت و سی نه در بافت چربی زیرپوستی ایجاد نمیکند؛ که احتمالا بیانگر این است که تمرین استقامتی مورد استفاده در این مطالعه، تغییری در ویژگی بافت چربی سفید از نظر قهوهای شدن یا سفید شدن بافت چربی سفید، ایجاد نمیکند.

مقایسۀ اثر فعالیت تناوبی شدید با حرکت همزمان، متوالی و متناوب بالاتنه و پایینتنه بر آیریزین وBDNF پلاسمای زنان چاق
هدف از پژوهش حاضر مقایسه اثر فعالیت تناوبی شدید با سه مدل چیدمان؛ همزمان، متوالی و متناوب فعالیت بالاتنه و پایینتنه بر آیریزین و BDNF پلاسمای زنان چاق بود. 11زن تمرین نکردهی چاق، 5/8±30/5 ساله و درصد چربی؛ 4/0±39/9، طی سه روز غیر متوالی، سه پروتکل روی دستگاه الپتیکال را با اعمال طرح متقاطع اجرا کردند. در پروتکل متوالی؛ پدال زدن دست، سپس پا بصورت متوالی هرکدام 10 تکرار با 90% واتحداکثر به مدت 40 دقیقه، در پروتکل همزمان؛10 تکرار پدال زدن دست با پا بصورت همزمان با 90% واتحداکثر به مدت 29 دقیقه و در پروتکل متناوب؛ پدال زدن دست و پا بصورت متناوب هر کدام 14 تکرار با 90% واتحداکثر به مدت 26 دقیقه و40 ثانیه، اجرا گردید. وهلههای یکدقیقهای فعالیت توسط استراحتهای فعال یکدقیقهای با 30% واتحداکثر تفکیک شدند. نمونهخونی قبل و یکساعت پس از اتمام پروتکلها اخذ شد. نتایج تحلیل واریانس با اندازههای تکراری و آزمون تعقیبی LSD، تفاوت معناداری (p≤0/05) در مقایسه تغییرات آیریزین و یا BDNF بین پروتکلها نشان نداد اما در پروتکلهای همزمان، متوالی و متناوب به ترتیب با افزایش 28/24 (0/02=p)، 21/86 و 17/18 (0/03=p) درصدی سطوح آیریزین و کاهش 0/01، 10/08 و 10/76 درصدی سطوح BDNF، نسبت به مقادیر پایه روبرو بودیم. در مجموع علیرغم صرف زمان کمتر در مدلهای همزمان و متناوب نسبت به مدل متوالی و درک فشار کمتر مدل متناوب نسبت به مدل همزمان، به نظر میرسد ترشح آیریزین و BDNF بیشتر متاثر از انرژی مصرفی طی ورزش باشد تا شدت، مدت و یا مدل چیدمان فعالیت عضلات.

در ارتباط با تأثیر ترکیب تمرینات ورزشی و مصرف الکل بر تنظیم کنندههای سیستم دفاع آنتی اکسیدانی به ویژه فاکتور هستهای اریتروئید 2 مرتبط با فاکتور2 (Nrf2) در عضله قلب مطالعات محدودی انجام شده است. هدف مطالعه حاضر، بررسی اثر هشت هفته تمرین هوازی توأم با مصرف اتانول بر بیان ژنNrf2 عضله قلب و شاخصهای آنتیاکسیدانی پلاسما در موش صحرایی نر بود. تعداد 32 سر موش صحرایی با میانگین وزنی6±230 گرم به چهار گروه کنترل، تمرین هوازی، اتانول20% با دوز 4 گرم بر کیلوگرم وزن بدن و اتانول به همراه تمرین تقسیم شدند. در پایان دوره سطوح میزان بیان ژن Nrf2 و میزان ظرفیت تام آنتیاکسیدانی (TCA) و مالون دیآلدئید (MDA) مورد ارزیابی قرار گرفتند. دادهها توسط آزمون تحلیل واریانس دو سویه و در سطح معنیداری 0.05≥p تجزیه و تحلیل شدند. نتایج نشان داد که تمرین هوازی اثر معنیداری در بیان ژن Nrf2 دارد (P=0.0068). نتایج عدم تأثیر معنیدار مصرف اتانول(P=0.312) و اثر تعاملی بین تمرین هوازی و مصرف اتانول (P=0.237) را در بیان ژن Nrf2 نشان دادند. تمرین هوازی بطور معنیداری بیان ژن Nrf2 را در گروههای تمرین و تمرین به همراه اتانول نسبت به گروه کنترل افزایش داد. مصرف اتانول بطور معنیداری سبب کاهش میزان TCA و افزایش MDA در مقایسه با سایر گروهها شد. یافتهها نشان داد که مصرف اتانول به ترتیب سبب کاهش و افزایش سطوح پلاسمایی TAC و MDA میگردد. در مقابل انجام تمرین هوازی از طریق افزایش سطوح TAC و افزایش بیان ژن Nrf2 منجر به کاهش آسیب اکسیداتیو ناشی از مصرف اتانول شد.

تأثیر تمرین شنا بر سلولهای گانگلیونیک و میزان بیان پروتئین NeuN در گانگلیون ریشۀ خلفی رتهای نر ویستار
یکی از نشانگرهای تمایز نرونی، پروتئین NeuN است که در نرونهای بالغ بیان میشود. هدف از پژوهش حاضر، بررسی اثر 20 روز تمرین استقامتی شنا بر پروتئین NeuN و تعداد سلولهای گانگلیونیک در گانگلیون ریشه خلفی موشهای صحرایی نر نژاد ویستار بود. در این پژوهش تجربی 16 رت با 6 هفته سن نر ویستار با وزن 250-200 گرم به عنوان نمونه پژوهش از مرکز نگهداری حیوانات آزمایشگاه پاستور تهیه و بهطور تصادفی به دو گروه (کنترل و تجربی) تقسیم شدند. گروه تجربی به مدت 20 روز فعالیت شنا (30 دقیقه در روز برابر با 65 درصد Vo2max) را انجام دادند. 48 ساعت بعد از آخرین جلسه تمرینی، حیوانات بیهوش و گانگلیون ریشه خلفی مناطق اندام تحتانی آنها جداسازی شد. برای ارزیابی جمعیت گانگلیونیک و پروتئین NeuN بهترتیب از تست کریزیل ویوله و ایمونوهیستوشیمی استفاده شد ، از آزمون آماری t مستقل برای تعیین تفاوت بین گروهها استفاده شد. نتایج پژوهش نشان داد که تمرین استقامتی شنا باعث افزایش معنیداری در بیان پروتئین NeuN میشود (p <0.05). علاوه بر این، تعداد سلولهای گانگلیونیک در گروه تمرین ورزشی استقامتی شنا افزایش معناداری را نشان داد (p <0.05). این مطالعه برای اولینبار نشان داد که تمرین شنا موجب افزایش تعداد سلولهای حسی و همچنین افزایش تمایز آنها و در نتیجه توسعه نرونهای حسی بالغ در گانگلیون ریشه خلفی نخاع میشود.

تأثیر هشت هفته تمرین مقاومتی، استقامتی و موازی بر مقادیر سرمی CTRP-12، فورین، KLF-15، پروفایل لیپیدی و مقاومت به انسولین در مردان چاق غیرفعال
آدیپوکاینها و بیومارکرهای التهابی، تسهیل کننده اثرات تخریبی چاقی نظیر مقاومت به انسولین هستند که در القای بیماری متابولیکی تقش بسزایی دارند. هدف پژوهش حاضر بررسی تاثیر ۸ هفته تمرین مقاومتی، استقامتی و موازی بر مقادیر سرمی CTRP-12، فورین، KLF-15 و مقاومت به انسولین در مردان چاق غیر فعال بود.40 مرد چاق غیرفعال (45ـ30 سال، BMI≥30) به طور تصادفی در 4 گروه، تمرین مقاومتی، تمرین استقامتی، تمرین موازی و کنترل تقسیمبندی شدند. گروههای تمرینی به مدت 8 هفته، سه جلسه در هفته به تمرینات ویژه پرداختند. مقادیر سرمی CTRP-12، فورین و KLF-15 با استفاده از کیت به روش ELISA اندازهگیری شد. برای تجزیه و تحلیل دادهها از آزمون آماری t همبسته و ANOVA در سطح معناداری (05/0 ≥p) استفاده شد. نتایج آزمون t همبسته نشان داد که مقادیر KLF-15 گروههای تمرین هوازی (p=0.006) و موازی (p=0.036) به طور معناداری افزایش یافت. در حالی که، مقادیر فورین در این دو گروه کاهش معناداری نشان داد (به ترتیب: p=0.001 p=0.044). مقادیر CTRP-12 نیز تنها در گروه تمرین موازی افزایش معناداری (p=0.023) داشت. تمرین هوازی (p=0.009) و موازی (p=0.006) به طور معناداری مقاومت به انسولین را کاهش دادند. در بررسی بین گروهی تمرین هوازی تغییرات معناداری در KLF-15 (p=0.039 و F=3.090) و فورین (p=0.031 و p=3.297) نسبت به گروه کنترل ایجاد کرد. به نظر میرسد استفاده از تمرینات هوازی و به ویژه تمرینات موازی تاثیرات بهتری بر پروفایل لیپیدی و همچنین آپریگولیشن آدیپوکاینهای ضد التهابی جدید نظیر KLF-15 و CTRP-12 و همچنین دانرگولیشن آدیپوکاین التهابی فورین داشته باشد.

تدوین و بهروزرسانی نُرمهای آمادگی جسمانی مردان و زنان 18 تا 60 سالۀ شهر کرمانشاه
هدف کلی این پژوهش تدوین و به روز رسانی نورمهای آمادگی جسمانی مردان و زنان 18 تا 60 سال شهر کرمانشاه بود. آزمودنیهای این پژوهش را 200 نفر از مردان و 200 نفر از زنان شهر کرمانشاه تشکیل دادند که به ترتیب، متوسط سن آنها 421/11±82/34 و 31/1±96/36 سال، قد 297/5±64/179 و57/0±46/161 سانتیمتر ، وزن 586/11±97/80 و 17/1±86/70 کیلوگرم بود که به طور تصادفی انتخاب و در طبقات مختلف سنی تقسیم شدند. از آزمونهای دویدن/ راه رفتن راکپورت برای اندازهگیری استقامت قلبی- عروقی، شنا برای سنجش استقامت عضلات کمربند شانهای، درازونشست برای سنجش استقامت عضلات ناحیة شکم، نیروسنج دستی برای سنجش قدرت عضلات مچ دست، جعبة اندازهگیری انعطاف بدنی برای تعیین انعطاف تنه، چین پوستی سه ناحیهای برای تخمین درصد چربی بدن استفاده شد. یافته های این پژوهش نشان داد که میانگین استقامت قلبی- عروقی آزمودنیها در مردان و زنان به ترتیب،14/45 و 80/38 میلی لیتر برکیلوگرم بر دقیقه، درصد چربی بدن 28/23 و 95/31 درصد، WHR 927/0 و 829/0 متر، انعطافپذیری 43/29 و 91/23 سانتیمتر، قدرت عضلانی پنجه دست راست 21/42 و 63/24 کیلوگرم، قدرت عضلانی پنجه دست چپ 02/36 و 13/23 کیلوگرم، استقامت عضلات سینهای تعداد 18/44 و 32/14 شنای سوئدی، استقامت عضلات شکم تعداد 81/32 و 94/28 دراز و نشست بود. در مقایسه با هنجارهای موجود، مردان شهر کرمانشاه در متغیرهای توان هوازی، قدرت عضلانی پنجه، استقامت عضلات سینه وضعیت بهتری به نسبت شهرهای دیگر و سایر کشورها داشتند و زنان شهر کرمانشاه در متغیرهای درصد چربی بدن، قدرت عضلات پنجه و انعطافپذیری وضعیت نامناسبی داشتند.

تأثیر چهار هفته تمرین هوازی بر بیان ژن فاکتور تغذیهای مشتقشده از سلولهای گلیال، TNF-α و عوامل شناختی در هیپوکمپ رتهای مبتلا به بیماری آلزایمر القاشده با آمیلویید بتا
زمینه و هدف : بیماری آلزایمر شایعترین شکل زوال عقل است و آمیلوئیدبتا نقشی در بیماری زایی آن ایفا میکنند. در سالهای اخیر ورزش منظم به عنوان یکی از سازوکارهای غیردارویی مهم برای مقابل با بیماری آلزایمر مطرح شده است. هدف از مطالعه بررسی تاثیر 4 هفته تمرین هوازی بر روی بیان ژن GDNF در رت های مبتلا به آلزایمر القا شده با Aβ1-42 بود. روش کار: آزمودنی های پژوهش شامل 56 سر رت نر ویستار 8 هفته ای با میانگین وزن 20±190 گرم بودند که به طور تصادفی به چهارگروه تقسیم شدند: گروه ورزش، گروه آلزایمر+ ورزش، گروه آلزایمر و گروه کنترل. Aβ1-42 به درون هیپوکمپ تزریق شد و 7 روز بعد از جراحی و توسعه بیماری آلزایمر رت های هر گروه به طور تصادفی قربانی شدند و گروهی تحت آزمون رفتاری قرار گرفتند. برای اندازه گیری بیان ژن GDNFبا استفاده از روش کمی Real time-PCR استفاده شد. نتایج: براساس مقایسه سطوح بیان ژن در چهار گروه مورد مطالعه، بیان ژن GDNF در بین گروههای تحقیق اختلاف معنی داری دارند بطوریکه ژن GDNF در گروه تمرین بیشترین و در گروه آمیلوئید بتا کمترین سطح بیان را داشته است (p <0.001). یادگیری و حافظه فضایی در گروه ورزش به طور معنی داری بهتر از گروه آلزایمری بود (p <0.001). نتیجه گیری: به نظر می رسد تمرین هوازی می تواند نقش بسزایی در بهبود یادگیری و همچنین افزایش بیان ژن های عامل تغذیه ای عصب در هیپوکمپ آلزایمری خصوصا GDNF داشته باشد که نهایتا به بهتر شدن حافظه و یادگیری کمک می کند.

تأثیر یک دوره تمرین ترکیبی و بازی در محیط غنیشده در دورۀ پیش از بلوغ بر ساختار هیپوکمپ موشهای صحرایی بزرگسال
حفظ ساختار و عملکرد مغز در طول زندگی از اهمیت بالایی برخوردار است. فرآیندهای کلیدی توسعه مغزی در دوره-های بحرانی رشد به ویژه در دوره پیش از بلوغ برجسته هستند. از این رو مداخلات محیطی در طی این دوره حساس رشدی، ممکن است به تغییرات قابل توجه و ماندگار در ساختارهای در حال رشد مانند هیپوکمپ منجر شوند. در این مطالعه، تاثیر تمرین ترکیبی و بازی در محیط غنی شده پیش از بلوغ بر ساختار هیپوکمپ موش های صحرایی بزرگسال مورد ارزیابی قرار گرفت. بدین منظور، تحقیق به صورت طولی انجام شد و 27 سر موش صحرایی نر پس از دوره شیردهی به سه گروه تمرین ترکیبی، بازی در محیط غنیشده و کنترل تقسیم شدند. مداخلات به مدت سه هفته در دوران قبل از بلوغ (48-28 روزگی) اجرا شدند و در ادامه تا دوره بزرگسالی (85-48 روزگی) همه حیوانات بدون مداخله درون قفس-های استاندارد نگهداری شدند. پس از این دوران، مراحل تشریح و بافتبرداری هیپوکمپ انجام شدند. روش وسترن بلات برای اندازهگیری عامل رشد عصبی مشتق از مغز (BDNF) و رنگآمیزی نیسل برای ارزیابی تعداد سلولها در نواحی هیپوکمپ استفاده شدند. نتایج آزمون آنوا یکطرفه نشان دادند که سطوح پروتئین BDNF (P
