Abstract
Aim of this study was to investigate the interactive effect of eight weeks of treadmill training and curcumin supplementation on the gene expression of pro-apoptotic factors in the skeletal muscle of aged rats with sarcopenia. In this experimental study, 30female Wistar rats were randomly distributed into five groups:healthy control, sarcopenia, sarcopenia+curcumin, sarcopenia+exercise, and sarcopenia + curcumin + exercise. The training protocol consisted of 15- 30 minutes of treadmill running at a speed of 20 meters per minute, five sessions per week for eight weeks.During the intervention period, the supplement groups were given 50 mg/kg curcumin daily by gavage. At the end of the intervention, body mass, soleus muscle, and the expression levels of Bax, Real-time PCR measured myostatin and caspase-3 genes in the soleus muscle tissue of the animals. The expression of Bax and caspase-3 genes in the healthy control group was significantly lower than in the other groups, and myostatin was significantly lower in the healthy control and sarcopenia+curcumin+training groups compared to the sarcopenia, sarcopenia+curcumin, and sarcopenia+training groups(p≥0.003).Caspase-3 and myostatin gene expression were higher in the sarcopenia group compared to other groups (p<0.017).In the sarcopenia+curcumin+exercise group, Bax gene expression was significantly reduced compared to sarcopenia, sarcopenia+curcumin, and sarcopenia+exercise groups (p≥0.036),and caspase-3 expression was significantly reduced compared to sarcopenia+ curcumin (p=0.015).According to these findings, aging is associated with an increase in markers related to cell death. Although aerobic exercise and curcumin supplementation have limited effects on pro-apoptotic factors, their simultaneous use is effective in modulating muscle tissue apoptosis by enhancing each other's effects
